Definition

The Two Hybrid Fragment Pooling Approach is used to identify the minimum functional regions required for protein interactions, mainly used in functional protein domain analysis and protein network construction. This method pairs a single bait with a random fragment prey pool, using degraded fragments to quickly locate the minimal protein region related to the interaction. Since multiple clones that encode overlapping protein regions can be identified, the initial screening visually shows the minimal domains required for the interaction.
