Neurotrophic agents are compounds that support neuronal growth, survival, and differentiation, playing a vital role in neural development and maintenance. Fellutamide A (15) demonstrates potent activity in stimulating nerve growth factor (NGF) synthesis and secretion in various neuronal and glial cell models, suggesting therapeutic potential for neurodegenerative disorders like Alzheimer's disease, with a distinct mechanism from epinephrine. 15G256α (50) enhances NGF-induced neurite outgrowth in PC12 cells at 100 μg/ml, although higher doses lead to cytotoxicity. Epolactaene (142) promotes neurite formation in SH-SY5Y cells, achieving 74% axon-bearing neurites at 10 μg/ml, approaching the efficacy of dibutyryl cAMP, indicating its neurogenic potential. Further in vivo studies are required to evaluate the therapeutic relevance of these agents.
