STING-IN-4 (Compound 1) is a STING inhibitor that inhibits STING expression and hence reducing activation of STING and nuclear factor-κB (NF-κB) signaling. STING-IN-4 shows anti-inflammatory activity and can be used for the research of sepsis.

STING[1]
STING-IN-4 (Compound 1) (20 μM; 26 h) inhibits NO production induced by LPS in RAW264.7 cells[1].
STING-IN-4 (2.5-10 μM; 26 h) significantly inhibits the expression of iNOS in RAW264.7 cells[1].
STING-IN-4 (5 and 50 μM; 12 h) significantly reduces STING degradation at 49, 52, and 55 °C and it may have an ability to interact with STING and enhance thermal stabilization of STING[1].
STING-IN-4 (2.5-10 μM; 8 h) inhibits LPS-induced activation of STING/IRF3/NF-κB[1].

Result:Significantly reduced the hemorrhage severity. Decreased the levels of alanine transaminase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) induced by LPS. Significantly reduced the levels of TNF-α, IL-6, and IFN-β compared with mice treated only with LPS. Markedly reduced the levels of STING, p-TBK, p-IRF3, p-p65, and p-IκB-α.

This protocol yields a clear solution of ≥ 3.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (35.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
