MGTA-117 Antibody

MGTA-117 is a humanized monoclonal antibody targeting CD117. MGTA-117 can be used for synthesis of antibody-drug conjugate (ADC), utilizing an amanitin payload. MGTA-117 has potent anti-tumor activity and increases survival in three acute myeloid leukemia (AML) xenograft hNSG mice models (Kasumi-1, AML PDX 1 and AML PDX 2). MGTA-117 enables hematopoietic stem cell transplantation (HSCT) preprocessing in AML, myelodysplasia with excess blasts (MDS-EB) and gene therapy.

In Vitro

MGTA-117 (0.1-1.0 mg/kg) rapidly and selectively binds and depletes CD117+ cells (human hematopoietic stem cells and AML tumor cells) with IC₅₀ s of <10 pM^[1].
MGTA-117 effectively depletes more than 95% of human hematopoietic stem and progenitor cells (HSPCs) in AML murine models^[3].

In Vivo

MGTA-117 (0.1 mg/kg, i.v., once for 21 days) has a short half-life with a t_1/2 of 91 h and selectively depletes ≥95% HSPCs in humanized NSG (hNSG) mice^[1,2].
MGTA-117(3 & 10 mg/kg, once or 3 mg/kg, QODx2) significantly increases survival of a 2.4-2.7-fold in Kasumi-1 xenograft hNSG mice model^[2].
MGTA-117 (3 & 10 mg/kg, once) significantly decreases peripheral tumor burden, delays tumor growth, and increases survival in AML PDX 1 (2-3.3 fold) and AML PDX 2 (1.3-1.8 fold) xenograft hNSG mice models^[2].
