Description

BRCA2 Protein, Human (HEK293, His, Strep, Flag) is the recombinant human-derived BRCA2, expressed by HEK293, with Strep, His, Flag labeled tag.

Background

BRCA2 is a protein involved in double-strand break repair and/or homologous recombination (HR). It binds RAD51 and enhances recombinational DNA repair by promoting RAD51 assembly onto single-stranded DNA (ssDNA). BRCA2 functions by targeting RAD51 to ssDNA over double-stranded DNA, enabling RAD51 to displace replication protein-A (RPA) from ssDNA and stabilizing RAD51-ssDNA filaments by blocking ATP hydrolysis. It is part of a PALB2-scaffolded HR complex containing RAD51C, which plays a role in HR-mediated DNA repair. BRCA2 may also participate in S phase checkpoint activation and selectively binds ssDNA, as well as ssDNA in tailed duplexes and replication fork structures. During the extension step after strand invasion at replication-dependent DNA double-strand breaks, BRCA2 collaborates with PALB2 in POLH localization at collapsed replication forks and DNA polymerization activity. Additionally, BRCA2 cooperates with NPM1 to regulate centrosome duplication. It interacts with the TREX-2 complex (transcription and export complex 2) subunits PCID2 and SEM1 and is required to prevent R-loop-associated DNA damage, thereby maintaining transcription-associated genomic stability.
