CKAP1/TBCB Protein, Human (His)

Cat. No.: HY-P77331
COA Handling Instructions

CKAP1/TBCB protein plays a pivotal role in the tubulin folding pathway, binding to alpha-tubulin folding intermediates after their chaperonin interaction, crucial for tubulin heterodimer transition. It regulates heterodimer dissociation, potentially acting as a negative regulator of axonal growth. The supercomplex, formed with cofactors A to E, captures and stabilizes tubulin, releasing committed polypeptides to the native state. CKAP1/TBCB's interactions with GAN and DCTN1 extend its functional roles in cellular processes. CKAP1/TBCB Protein, Human (His) is the recombinant human-derived CKAP1/TBCB protein, expressed by E. coli, with N-His labeled tag. The total length of CKAP1/TBCB Protein, Human (His) is 244 a.a., with molecular weight of 33-37 kDa.

Background

The CKAP1/TBCB protein exhibits a multifaceted role in the tubulin folding pathway, binding to alpha-tubulin folding intermediates subsequent to their interaction with cytosolic chaperonin, a pivotal step in the transition from newly synthesized tubulin to properly folded heterodimer. It plays a crucial role in regulating tubulin heterodimer dissociation and may function as a negative regulator of axonal growth. The formation of a supercomplex composed of cofactors A to E is integral to its mechanism. Cofactors A and D function by capturing and stabilizing tubulin in a quasi-native conformation, while cofactor E binds to the cofactor D-tubulin complex. The subsequent interaction with cofactor C leads to the release of tubulin polypeptides committed to the native state. Additionally, cofactors B and E can form a heterodimer that binds to alpha-tubulin, enhancing their capacity to dissociate tubulin heterodimers. CKAP1/TBCB also interacts with GAN and DCTN1, further expanding its functional associations in cellular processes.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
