CYP7A1 Protein, Human (His) is the recombinant human-derived CYP7A1, expressed by E. coli , with N-6*His labeled tag.
Background
CYP7A1 is a cytochrome P450 monooxygenase involved in the metabolism of endogenous cholesterol and its oxygenated derivatives (oxysterols). It utilizes molecular oxygen to insert one oxygen atom into a substrate while reducing the second into a water molecule, with two electrons supplied by NADPH via cytochrome P450 reductase (CPR; NADPH-ferrihemoprotein reductase). As a critical regulatory enzyme in bile acid biosynthesis and cholesterol homeostasis, CYP7A1 catalyzes the hydroxylation of the 7-alpha carbon-hydrogen bond of cholesterol, a rate-limiting step in cholesterol catabolism and bile acid synthesis. Additionally, it 7-alpha hydroxylates various oxysterols, including 4beta-hydroxycholesterol and 24-hydroxycholesterol, and oxidizes the 7,8 double bond of 7-dehydrocholesterol and lathosterol, predominantly forming 7-keto derivatives.
