Background
DGCR8 is a component of the microprocessor complex that functions as an RNA- and heme-binding protein involved in the initial step of microRNA (miRNA) biogenesis. Within the microprocessor complex, DGCR8 processes primary miRNA transcripts (pri-miRNAs) into precursor miRNAs (pre-miRNAs). It acts as a molecular anchor to recognize the dsRNA-ssRNA junction of pri-miRNAs and directs DROSHA to cleave 11 bp away from the junction, releasing hairpin-shaped pre-miRNAs for subsequent processing by cytoplasmic DICER into mature miRNAs. The heme-bound DGCR8 dimer binds pri-miRNAs cooperatively as a trimer (of dimers) to trigger cleavage, while the heme-free monomer binds as a dimer with reduced activity. Both double-stranded and single-stranded regions of pri-miRNAs are required for binding. DGCR8 specifically recognizes and binds N6-methyladenosine (m6A)-modified pri-miRNAs, a key modification for processing. It is also involved in silencing embryonic stem cell self-renewal (by similar) and plays a role in DNA repair by promoting RNF168 recruitment to RNF8 and MDC1 at double-strand breaks for DNA damage clearance.
