Background
HuR is an RNA-binding protein that binds to the 3'-UTR of mRNAs to enhance their stability. It plays a role in embryonic stem cell (ESC) differentiation by preferentially stabilizing non-m6A-methylated mRNAs, thereby promoting ESC differentiation (By similarity). It can also bind m6A-containing mRNAs, such as MYC mRNA, contributing to MYC stability. HuR targets poly-U and AU-rich elements (AREs) in the 3'-UTR of mRNAs, including those in FOS and IL3/interleukin-3 mRNAs. For FOS, it binds a 27-nucleotide core ARE containing AUUUA, AUUUUA, and AUUUUUA motifs, with a preference for the 5'-UUUU[AG]UUU-3' motif in vitro. In collaboration with ZNF385A, HuR binds the 3'-UTR of p53/TP53 mRNA to regulate its CDKN2A-induced nuclear export, potentially modulating p53/TP53 expression and partially mediating CDKN2A's anti-proliferative effects (By similarity). Additionally, it stabilizes leptin mRNA containing an ARE in its 3'-UTR.
