Background

The FOLR2 protein plays a pivotal role in binding to folate and reduced folic acid derivatives, facilitating the transport of 5-methyltetrahydrofolate and folate analogs into the intracellular space. It exhibits a remarkable affinity for folate and folic acid analogs under neutral pH conditions. However, upon receptor endocytosis, exposure to a slightly acidic pH induces a conformational change in FOLR2, leading to a substantial reduction in its affinity for folates and subsequent release from the receptor, as observed in similar proteins.
