Background

Endoplasmin is an ATP-dependent chaperone involved in protein processing and transport regulation within the endoplasmic reticulum. It acts alongside MESD to modulate the Wnt pathway by facilitating the folding of LRP6, a coreceptor of the canonical Wnt pathway. When associated with CNPY3, Endoplasmin is essential for the proper folding of Toll-like receptors and promotes the folding and trafficking of TLR4 to the cell surface. Additionally, it may participate in the unfolding of leaderless cytosolic cargos (e.g., interleukin 1/IL-1) to enable their translocation into the ERGIC (endoplasmic reticulum-Golgi intermediate compartment) and secretion, mediated by the cargo receptor TMED10. by similar: cooperates with MESD in Wnt pathway regulation; interacts with CNPY3 for TLR folding; facilitates secretion of leaderless proteins via TMED10.
