Background
NLRP2 suppresses TNF- and CD40-induced NFKB1 activity at the IKK complex level by inhibiting TNF-triggered NFKBIA degradation. When bound to PYCARD, it activates CASP1, promoting the secretion of mature pro-inflammatory cytokine IL1B. NLRP2 is likely a component of the inflammasome, a protein complex also containing PYCARD, CARD8, and CASP1, which functions to activate pro-inflammatory caspases. by similar: NLRP2 shares mechanistic similarities with other inflammasome components (e.g., NLRP3) in regulating caspase activation and cytokine release during inflammatory responses.
