PLCD4 is a phospholipase that hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate two second messenger molecules, diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). DAG mediates the activation of protein kinase C (PKC), while IP3 triggers the release of Ca2+ from intracellular stores. During fertilization, PLCD4 is essential for the acrosome reaction in sperm, likely by serving as a key enzyme for intracellular Ca2+ mobilization in the zona pellucida-induced acrosome reaction. It may also play a role in cell growth and collaborates with nuclear PKC to modulate liver regeneration. Overexpression of PLCD4 upregulates the Erk signaling pathway and promotes cell proliferation. by similar: Other phospholipases may also participate in similar PIP2 hydrolysis and signaling processes.
