Background

Rab10, a member of the Rab small GTPase family, serves as a key regulator of intracellular membrane trafficking, governing vesicle formation, movement, tethering, and fusion (By similarity). It primarily mediates biosynthetic protein transport from the Golgi to the plasma membrane (By similarity), exemplified by its role in delivering SLC2A4/GLUT4 glucose transporter-enriched vesicles to the plasma membrane (PubMed:17403373, PubMed:22908308, PubMed:27354378). Rab10 also facilitates TLR4 (a toll-like receptor) trafficking to the plasma membrane, potentially influencing innate immunity (PubMed:20643919). In neurons, it promotes axonogenesis via vesicular transport regulation toward axonal plasma membranes (By similarity), while in epithelial cells, it directs Golgi-to-basolateral membrane transport (By similarity). Additional roles include participation in the basolateral recycling pathway, phagosome maturation (By similarity), and modulation of endoplasmic reticulum dynamics (By similarity). Collaborating with LRRK2, RAB8A, and RILPL1, Rab10 regulates ciliogenesis (By similarity); when phosphorylated by LRRK2 at Thr-73, it binds RILPL1 to inhibit cilia formation (By similarity). It contributes to protein export via a Rab8-Rab10-Rab11-dependent endosomal pathway (By similarity) and, upon LRRK2-mediated phosphorylation, stabilizes on stressed lysosomes to drive lysosomal content release through EHBP1/EHNP1L1 effectors (PubMed:30209220).
