Background

SIGMAR1 functions in lipid transport from the endoplasmic reticulum and is involved in a wide array of cellular functions, likely through regulating the biogenesis of lipid microdomains at the plasma membrane. It modulates various receptors, playing roles in BDNF signaling and EGF signaling, and regulates ion channels such as the potassium channel, potentially influencing neurotransmitter release. Additionally, SIGMAR1, together with ANK2, modulates ITP3R-dependent calcium efflux, contributing to calcium signaling. It is implicated in diverse cellular processes including proliferation, survival, and death. Initially identified for its binding to psychoactive drugs, SIGMAR1 participates in learning, memory, and mood regulation. In motor neurons, it is essential for proper mitochondrial axonal transport, particularly the retrograde movement of mitochondria. Through its interaction with RNF112, SIGMAR1 also helps protect cells against oxidative stress-induced cell death (by similarity).
