SLC5A8 functions as an electrogenic sodium (Na(+)) and chloride (Cl-)-dependent sodium-coupled solute transporter, facilitating the transport of monocarboxylates (including short-chain fatty acids such as L-lactate, D-lactate, pyruvate, acetate, propionate, valerate, and butyrate), monocarboxylate drugs (e.g., nicotinate, benzoate, salicylate, and 5-aminosalicylate), and ketone bodies (e.g., beta-D-hydroxybutyrate, acetoacetate, and alpha-ketoisocaproate), with a Na(+):substrate stoichiometry ranging from 4:1 to 2:1. It also catalyzes passive carrier-mediated diffusion of iodide, mediates iodide transport from thyrocytes into the colloid lumen through the apical membrane, and may be involved in the intestinal absorption of D-lactate and monocarboxylate drugs. As a tumor suppressor, SLC5A8 inhibits colony formation in colon cancer, prostate cancer, and glioma cell lines. It may play a critical role in the neuronal uptake of L-lactate and ketone bodies via an electrochemical Na(+) gradient-driven process, supporting neuronal energy metabolism and function. Additionally, SLC5A8 mediates sodium-coupled electrogenic transport of pyroglutamate (5-oxo-L-proline) and can transport chloride, bromide, iodide, and nitrate ions under reduced external sodium ion concentrations.
