Background

SPOP is a component of the cullin-RING-based BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex that mediates target protein ubiquitination, often leading to proteasomal degradation. In complex with CUL3, it ubiquitinates and degrades BRMS1, DAXX, PDX1/IPF1, GLI2, and GLI3, while ubiquitinating MACROH2A1 and BMI1 without degradation. It suppresses PDX1/IPF1 target gene (e.g., insulin) activation by promoting PDX1/IPF1 degradation. The homodimeric SPOP-containing BCR complex exhibits higher ubiquitin ligase activity than the SPOP/SPOPL heterodimer-containing complex. It regulates BET proteins (BRD2/3/4) stability, controls translation (not degradation) of DNA replication factors CDT1 and CDC6 for DNA synthesis, targets IRF1 via S/T-rich degrons, restricts EV71 replication by lysosomal degradation of viral protease 2A, and inhibits HBV via HNF1A ubiquitination. Additionally (by similar), it ubiquitinates BRDT to regulate histone removal during spermatogenesis.
