  • Function

    KCNQ4 is the pore-forming subunit of the voltage-gated potassium (Kv) channel that regulates the excitability of sensory cells in the cochlea. The KCNQ4/Kv7.4 channel consists of four pore-forming subunits assembled as tetramers, facilitating potassium ion efflux during the repolarization phase of action potentials to modulate membrane potential in excitable cells. This channel conducts slowly activating and deactivating currents, often exhibiting inward rectification at positive potentials. In vitro studies suggest selectivity for other cations beyond potassium, with affinity ranking K(+) = Rb(+) > Cs(+) > Na(+). KCNQ4 is critical for normal inner ear functions, including sound perception.

    Interacts (via C-terminus) with calmodulin; forms a heterooctameric structure (with 4:4 KCNQ1:CALM stoichiometry); the interaction is calcium-independent, constitutive, participates in the proper assembly of a functional channel. The interaction with calcium-free CALM controls channel trafficking whereas interaction with calcium-bound CALM regulates channel gating. May form a functional heteromultimeric channel with KCNQ3. Interacts with HSP90AB1; promotes cell surface expression of KCNQ4.
