Background

Function
CD41 (Integrin αIIb, GPIIb) is an integrin α subunit encoded by the ITGA2B gene. Together with CD61 (Integrin β3), it forms αIIbβ3 integrin (GPIIb/IIIa), which is a crucial receptor in platelet aggregation and hemostasis. CD41 is mainly expressed in platelets and megakaryocytes, mediating platelet aggregation and stabilizing thrombus formation by binding ligands such as fibrinogen and vWF. CD41 is not only a classic biomarker for platelets and megakaryocytes, but also an important target for immunophenotyping of acute megakaryocytic leukemia (AML-M7), hematopoietic development research, and the development of antiplatelet drugs.

Expression
Tissue_specificity:Isomers 1 and 2 are expressed in platelets and megakaryocytes, but not in reticulocytes. They were not detected in Jurkat cells or the U937 cell line (PubMed: 2351656) . Isomer 3 is expressed in prostate adenocarcinoma and various erythroleukemia, prostate adenocarcinoma, and melanoma cell lines, including PC-3, DU-145, HEL, WM983A, WM983B, and WM35. It was not detected in platelets or normal prostate tissue (protein level) (PubMed: 9809974) . -
Isoforms & Post-Translational Modification
CD41 has 3 isomers: P08514-1: 113377 Da (predicted); P08514-2: 109574 Da (predicted); P08514-3: 103218 Da (predicted).
Cleaved by ELANE; the cleavage promotes activation of platelet fibrinogen receptor integrin alpha-IIb/beta-3 -
Subunit
Heterodimer of an alpha and a beta subunit. The alpha subunit is composed of a heavy and a light chain linked by a disulfide bond.
