Function
S100A12 is a calcium-binding S100/calgranulin protein secreted by activated neutrophils and granulocytes, with proinflammatory activity centered on interaction with RAGE[1][2]. Mechanistically, S100A12-RAGE engagement activates NF-κB-dependent endothelial and inflammatory signaling, supporting cytokine production, adhesion biology, and chronic cellular activation[2][3]. In disease models and human inflammatory samples, S100A12 is strongly expressed in active inflammatory bowel disease and inflamed synovial tissue, and circulating levels correlate with disease activity[1][2]. Compared with related S100/calgranulin isoforms, S100A12 has a key experimental distinction because functional S100A12 is absent from the murine genome, limiting direct interpretation of standard mouse models[4]. For experimental applications, structural studies show that S100A12 binds the RAGE V domain, while tranilast and FPS-ZM1 reduced S100A12-induced cellular responses in vitro[5].
