Apovincaminic acid hydrochloride salt
Based on 1 publication(s) in Google Scholar
Apovincaminic acid hydrochloride salt is the major metabolite of Vinpocetine (HY-13295), with blood-brain barrier permeability and oral activity. Apovincaminic acid hydrochloride salt is detectable in the plasma of pregnant rabbits after oral administration of Vinpocetine. Apovincaminic acid hydrochloride salt is absorbable from all segments of the gastrointestinal tract of Norway rat, with the stomach as the primary absorption site, and is excreted via urine and feces. Apovincaminic acid hydrochloride salt is used in the research of cerebrovascular diseases.

Description
In Vitro
Apovincaminic acid hydrochloride salt is the major metabolite of Vinpocetine (HY-13295), a vinca alkaloid derivative that blocks Na+ channels. Vinpocetine inhibits IKK with an IC50 value of 17.17 μM. Vinpocetine also acts as a PDE inhibitor and suppresses NF-κB-dependent inflammatory responses by directly targeting IKK[1][2].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
In Vivo
Apovincaminic acid (0.148 μCi, 10 mg/kg; direct injection into ligated gastrointestinal segments, i.v., p.o.; 1 hour, 72 hours) hydrochloride salt is absorbed from all tested segments of the Rattus norvegicus gastrointestinal tract (with notable uptake in stomach and jejunum walls), with approximately 50% of an oral dose absorbed, and is excreted primarily in urine after i.v. administration and in faeces after oral administration[2].

Animal Model:RG-Wistar (male, 180-210 g)[2]
-
Dosage:0.148 μCi (in situ study); 10 mg/kg (excretion study)
-
Administration:direct injection into ligated gastrointestinal segments; 1 hour (in situ study); i.v., p.o.; 72 hours (excretion study)
-
Result:Absorbed Apovincaminic acid from all tested gastrointestinal segments.
Retained most drug dose in stomach, duodenum and jejunum lumen and wall 1 h after oral intake.
Recovered total 79.26% radioactive substance in urine and feces within 72 h post intravenous injection.
Recovered total 80.19% radioactive substance in urine and feces within 72 h post oral administration.
Revealed about 50% absorption rate for oral dosage form.
Detected original apovincaminic acid in urine under both administration modes.
