Carboxy SNARF-1, acetoxymethyl ester (5/6-mixture) is a fluorescent probe used for intracellular pH measurement. The detection wavelengths of Carboxy SNARF, acetoxymethyl ester are Ex/Em = 540/590 nm and 540/650 nm, with an excitation wavelength of 540 nm. Emission signals at 590 nm and 650 nm are recorded simultaneously, and the ratio of these emission signals yields a linear measurement of intracellular pH.

2. Working Solution Preparation
2.2 Working concentration: 5 μM.
2.3 Note: Adjust working solution concentration as needed; prepare fresh before use.
3. Staining Procedure
3.1 Sample type: Isolated rat portal vein smooth muscle tissue strips.[1]
3.2 Incubation conditions: Incubate tissue strips with 5 μM carboxy SNARF, acetoxymethyl ester for 1 h at room temperature.
5. Detection & Analysis
5.1 Instrument: Inverted fluorescence microscope.
5.1.1 Excitation wavelength: 540 nm; emission wavelengths: 590 nm and 650 nm.
5.2 Result analysis:
5.2.1 Calculate the ratio of emission signals at 590 nm and 650 nm to obtain linear intracellular pH measurements.
5.2.2 Calibrate pH measurements using either nigericin or an in vitro mock intracellular solution with known pH values.
5.2.3 Stable fluorescence recordings for intracellular pH measurement can be obtained for 1-2 h following loading.
