Endoglycoceramidase II (EGCase II) is an endo-β-glucosidase that releases intact glycans from ceramides in glycosphingolipids. Endoglycoceramidase II catalyzes the hydrolysis of β-glycosidic linkages between oligosaccharides and ceramides in various glycosphingolipids[1].

In Vitro

Purified recombinant Endoglycoceramidase II (EGCase II) from Rhodococcus sp. (153 nM; 1 h) exhibits optimal enzymatic activity at pH 5.5, hydrolyzing 4MU-Lac in a time-dependent manner[1].
Purified recombinant Endoglycoceramidase II (EGCase II) from Rhodococcus sp. (5 μM) is thermolabile, with an apparent melting temperature of 42 °C at a 1 °C/min heating rate, a half-life of ~10 min at 37 °C, and kinetic stability at 25 °C[1].
Purified recombinant Endoglycoceramidase II (EGCase II) from Rhodococcus sp. is susceptible to tryptic digestion at 25 °C and 37 °C, with cleavage at Arg321/Arg332, but becomes resistant to digestion in the presence of lauryl glucoside or Triton X-100[1].
