Biological Activity
Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Preparation of FD-1080 Maleimide working solution
1.1 Preparation of stock solution
Dilute FD-1080 Maleimide with DMSO to prepare FD-1080 Maleimide stock solution.
Note: Aliquot the FD-1080 Maleimide stock solution and store at -20℃ or -80 ℃ protected from light.
1.2 Preparation of working solution
Dilute the stock solution with pre-warmed PBS to prepare 2-10 μM FD-1080 Maleimide working solution. Sterilize by filtration through a 0.2 μm filter membrane.
Note: Adjust the concentration of FD-1080 Maleimide working solution according to actual conditions and prepare fresh before use.
2. Cell Staining
2.1 Cell preparation
Suspension cells: Collect cells by centrifugation and wash twice with PBS for 5 minutes each time.
Adherent cells: Discard the culture medium and digest cells with trypsin. After centrifugation, discard the supernatant and wash twice with PBS for 5 minutes each time.
2.2 Add 1 mL FD-1080 Maleimide working solution and incubate at room temperature for 10-30 minutes.
2.3 Centrifuge at 400 g, 4℃ for 3-4 minutes and discard the supernatant.
2.4 Wash cells twice with PBS for 5 minutes each time.
2.5 Resuspend cells in 1 mL serum-free medium or PBS, then detect under a fluorescence microscope or flow cytometer.
3. In vivo injection
3.1 Inject 200 μL of 80 μM FD-1080 Maleimide working solution intravenously into mice, and perform in vivo imaging analysis after 10-20 minutes.
