Background
Beta-Catenin protein functions as a key downstream component in the canonical Wnt signaling pathway, regulating various cellular processes. In the absence of Wnt, it forms a complex with AXIN1, AXIN2, APC, CSNK1A1, and GSK3B, promoting phosphorylation and ubiquitination of CTNNB1 for subsequent degradation. Upon Wnt ligand binding, CTNNB1 accumulates in the nucleus, acting as a coactivator for transcription factors of the TCF/LEF family to activate Wnt-responsive genes. Beyond its role in Wnt signaling, beta-Catenin is involved in cell adhesion, acting as part of an E-cadherin:catenin adhesion complex. It plays a role in regulating centrosome cohesion, CDK2/PTPN6/CTNNB1/CEACAM1-mediated insulin internalization, and promotes neurogenesis. Additionally, beta-Catenin interacts with various proteins, including AXIN1, SRC, TCF7L2, and others, highlighting its involvement in intricate molecular networks governing diverse cellular functions. The dynamic interactions and dual roles of beta-Catenin underscore its significance in cellular homeostasis and signaling cascades.
