CENPE is a microtubule plus-end-directed kinetochore motor protein that plays pivotal roles in chromosome congression, microtubule-kinetochore conjugation, and spindle assembly checkpoint (SAC) activation. It drives chromosome congression (metaphase plate formation) by mediating lateral sliding of polar chromosomes along spindle microtubules toward the equator and facilitating kinetochore-microtubule attachment stabilization. Dettyrosinated microtubule tracks preferentially transport pole-proximal chromosomes. As a processive bi-directional tracker of dynamic microtubule tips, CENPE persistently strengthens kinetochore-microtubule linkages post-congression. It suppresses congression in NDC80-depleted cells and promotes it only when microtubules are stabilized. CENPE is essential for stable kinetochore-microtubule attachments, requiring its dependent recruitment of BUB1B, MAD1, and MAD2 to kinetochores. Through interaction with BUB1B, it activates SAC kinase activity to prevent aneuploidy from single chromosome loss (by similar).
