Background
CLC1 is a voltage-gated chloride channel that plays a key role in skeletal muscle excitability. It generates the majority of plasma membrane chloride conductance in skeletal muscle fibers, stabilizing the resting membrane potential and contributing to the repolarization phase during action potential firing. The channel forms a homodimer with each subunit possessing its own ion conduction pathway, producing double-barreled currents regulated by two types of gates: fast glutamate gates controlling each subunit independently and a slow common gate operating both subunits simultaneously. CLC1 exhibits significant open probability at resting membrane potential and is further activated upon depolarization. It shows ion selectivity for chloride > thiocyanate > bromide > nitrate > iodide.
