Properdin is a crucial positive regulator within the alternate pathway (AP) of complement activation, playing a pivotal role in enhancing the stability of the C3- and C5-convertase enzyme complexes. By binding to these complexes, Properdin inhibits the CFI-CFH mediated degradation of Complement C3 beta chain (C3b). In its plasma form, Properdin forms dimers, trimers, or tetramers in specific proportions. Notably, Properdin interacts with the pro-C3-convertase enzyme complex (C3b-Bb), consisting of Complement C3 beta chain (C3b) and the Complement factor B Bb fragment (Bb), through its TSP type-1 5 domain. This interaction is pivotal for stabilizing the complex, transforming it into the active C3-convertase enzyme complex (C3b-Bb-FP). Furthermore, Properdin interacts directly with C3b and Complement factor B (CFB), highlighting its multifaceted role in complement activation and regulation.
