Background
DDX21, a member of the DEAD-box subfamily, is a multi-tasking RNA helicase that utilizes energy from ATP hydrolysis to the unwinding of structured RNA molecules, thereby enabling downstream RNA processing or remodeling of RNA-rotein complexes. DDX21 binds to various RNA substrates, such as rRNA, snoRNA, 7SK, and nucleic acid structures, including R-loops that are often formed co-transcriptionally by nascent RNA hybridized with template DNA. DDX21 has been implicated in a wide range of biological processes, including multiple steps in ribosome biogenesis, regulation of RNA polymerase II (RNAPII)-mediated transcription, and sensing of cellular glucose levels for epidermal differentiation, among others. These multifaceted functions have been associated with many developmental defects and diseases, including cancers, possibly reflecting the fact that DDX21 may reside in diverse complexes to mediate corresponding cellular processes[1].
