The EZH2 protein is a member of the Polycomb group (PcG). EZH2 Protein, Human (sf9, His) is the recombinant human-derived EZH2, expressed by Sf9 insect cells, with C-10*His labeled tag.

Background

EZH2, a Polycomb group (PcG) protein, acts as the catalytic subunit of the PRC2/EED-EZH2 complex, contributing to histone methylation and subsequent transcriptional repression. This complex mono-, di-, and trimethylates 'Lys-27' (H3K27) of histone H3, generating H3K27me1, H3K27me2, and H3K27me3, respectively. EZH2 exhibits a preference for substrates with less methylation and is pivotal for forming H3K27me3 in embryonic stem cells, playing a crucial role in maintaining stem cell identity and facilitating proper differentiation. Moreover, the PRC2/EED-EZH2 complex may serve as a platform for recruiting DNA methyltransferases, establishing a connection between distinct epigenetic repression systems. Target genes repressed by this complex include HOXC8, HOXA9, MYT1, CDKN2A, and retinoic acid-responsive genes. EZH2 extends its methylating activity beyond histones to non-histone proteins like the transcription factor GATA4 and nuclear receptor RORA. Additionally, EZH2 regulates the circadian clock through histone methylation at circadian gene promoters, contributing to CRY1/2-mediated repression of CLOCK-BMAL1-driven transcriptional activation of PER1/2. This multifaceted involvement underscores EZH2's integral role in epigenetic regulation and cellular processes.
