Background
eRF3a is the GTPase component of the eRF1-eRF3-GTP ternary complex, which mediates translation termination in response to stop codons UAA, UAG, and UGA. GSPT1/ERF3A delivers ETF1/ERF1 to stop codons: The eRF1-eRF3-GTP complex binds to a stop codon in the ribosomal A-site, and GTP hydrolysis by GSPT1/ERF3A induces a conformational change leading to its dissociation, allowing ETF1/ERF1 to fully occupy the A-site. Additionally, eRF3a is part of the transient SURF complex that recruits UPF1 to stalled ribosomes during nonsense-mediated decay (NMD) of mRNAs containing premature stop codons. It is also required for SHFL-mediated translation termination, which inhibits programmed ribosomal frameshifting (-1PRF) in viral and cellular mRNAs. by similar: Its functions are supported by studies on complex binding, conformational dynamics, and roles in NMD and frameshifting regulation.
