Background

HACE1, an E3 ubiquitin-protein ligase, intricately participates in Golgi membrane fusion and the regulation of small GTPases. Its crucial role in Golgi membrane dynamics during the cell cycle involves recruitment to the Golgi membrane by Rab proteins, where it orchestrates postmitotic Golgi membrane fusion. During mitotic Golgi disassembly, HACE1 mediates ubiquitination, serving as a pivotal signal for Golgi reassembly in the subsequent stages of cell division. Notably, HACE1 exhibits specificity in its interaction with GTP-bound RAC1, leading to the ubiquitination and subsequent degradation of active RAC1.
