JAG2 also promoted the migration and invasion of colon cancer cells in a non-EMT pathway[1][2]. Background
Modulation of JAG2 expression significantly affected in vivo tumorigenicity of CRC cell lines. Moreover, knockdown of JAG2 sensitized CRC cells to chemotherapeutic agents, while ectopic expression of JAG2 increased chemoresistance of the CRC cells. Significant down-regulation of p21 was observed in JAG2-knockdown cells. NOTCH signaling consists of four receptors (NOTCH1-NOTCH4), three delta-like ligands (DLL1, DLL3, and DLL4), and two serrate-like ligands (JAG1 and JAG2). Knockdown or pharmacological inhibition of β-catenin reduces JAG2 levels in the CRC cell lines[1].The human JAG2 gene is located on chromosome 14q32 and consists of 26 exons which encode mRNA variants with 5825 bp and 5721 bp in length. The protein encoded by this gene contains a signal peptide, an extracellular DSL region, 16 EGF-like repeats, a cysteine-rich region, a transmembrane region, and a short cytoplasmic region of 132 amino acids[2].
Verified Bioactivity
Measured by the ability of the immobilized protein to enhance BMP-2 induced alkaline phosphatase activity in C3H10T1/2 mouse embryonic fibroblast cells. The ED50 for this effect is 0.5-2 µg/mL.
