Background
KDM1B is a histone demethylase that specifically removes mono- and di-methyl groups from 'Lys-4' of histone H3 (without affecting tri-methylated state), functioning as a transcriptional corepressor. The enzyme utilizes FAD-dependent oxidation to generate an imine intermediate followed by hydrolysis. During oogenesis, KDM1B is required for de novo DNA methylation of specific imprinted genes. Biochemical studies demonstrate its remarkable selectivity: it shows no activity toward any methylation states of histone H3 'Lys-9', 'Lys-27', 'Lys-36', or histone H4 'Lys-20'.
