Background
Mitochondrial trans-2-enoyl-CoA reductase (MECR) is an oxidoreductase that catalyzes the last step in mitochondrial fatty acid synthesis (mtFAS). MECR catalyzes the NADPH-dependent reduction of trans-2-enoyl thioesters in mitochondrial fatty acid synthesis (fatty acid synthesis type II). Fatty acid chain elongation in mitochondria uses acyl carrier protein (ACP) as an acyl group carrier, but MECR accepts both ACP and CoA thioesters as substrates in vitro. MECR provides octanoyl chain for lipoic acid biosynthesis, regulating protein lipoylation and mitochondrial respiratory activity. Knockdown of MECR inhibits cell proliferation and colony formation, promotes apoptosis, and inhibits metastasis in HCC cell lines BEL-7404. In addition, MECR mutations cause childhood-onset dystonia and optic atrophy[1][2][3][4][5][6].
