Background

nsp3 (papain-like protease) is a key functional protein in the viral replicase polyprotein, mainly responsible for the cleavage of the N-terminus of the replicase polyprotein; it participates in the assembly of virus-induced cytoplasmic double-membrane vesicles (DMVs) together with nsp4 (DMVs are necessary for viral replication) and plays a role in its formation and maintenance (DMVs are formed by nsp3 and nsp4, and nsp6 is involved in the connection of the endoplasmic reticulum membrane and the binding to lipid droplets). At the same time, nsp3 inhibits the induction of type I interferon by blocking the phosphorylation, dimerization and nuclear translocation of host IRF3, and can also inhibit NF-κB signaling, thereby antagonizing innate immunity. In addition, it has deubiquitination/de-ISG activity, can process Lys-48 and Lys-63-linked polyubiquitin chains in cellular substrates, preferentially cuts ISG15 from the antiviral protein IFIH1 (MDA5) (has no effect on RIGI), and participates in the host ADP-ribosylation process.
