Background
Netrin-1, a crucial regulator of axonal guidance in both the central nervous system (CNS) and peripheral motor neurons, plays a pivotal role in directing commissural axons within the CNS and motor axons in the periphery. The association of Netrin-1 with either DCC or certain UNC5 receptors dictates whether axons are attracted or repelled. Specifically, binding to UNC5C induces the dissociation of UNC5C from polymerized TUBB3 in microtubules, leading to heightened microtubule dynamics and subsequent axon repulsion. Netrin-1 is essential for dorsal root ganglion axon projection toward the spinal cord and functions as a survival factor by preventing the initiation of apoptosis through its interactions with receptors. Furthermore, Netrin-1's involvement in tumorigenesis underscores its regulatory role in apoptosis. The protein binds to a repertoire of receptors, including DCC, UNC5A, UNC5B, UNC5C, UNC5D, and DSCAM, and exhibits interaction with APP, highlighting its multifaceted roles in cellular processes.
