Biological Activity

PLK4 is a serine/threonine-protein kinase that plays a central role in centriole duplication by triggering procentriole formation on parental centrioles and recruiting biogenesis proteins like SASS6, CPAP, and gamma-tubulin. Overexpression induces centrosome amplification via multiple procentriole generation during S phase. It phosphorylates FBXW5 at Ser-151 to suppress SASS6 ubiquitination and regulates G1/S transition. Dysregulation may contribute to tumorigenesis due to frequent centrosome aberrations in tumors. PLK4 also mediates deuterosome-dependent centriole amplification (producing >100 centrioles) in multiciliated cells and promotes trophoblast differentiation by phosphorylating HAND1. Additional substrates include CDC25C, CHEK2, CEP131 (Ser-78), and PCM1 (Ser-372), the latter being critical for centriolar satellite integrity. by similar: PLK4 is required for STIL recruitment and STIL-driven centriole amplification.
