PRKACB is a protein kinase that mediates cAMP-dependent signaling triggered by receptor binding to GPCRs. PKA activation regulates diverse cellular processes such as cell proliferation, the cell cycle, differentiation, microtubule dynamics, chromatin condensation and decondensation, nuclear envelope disassembly and reassembly, as well as intracellular transport mechanisms and ion flux. PRKACB controls the abundance of compartmentalized pools of its regulatory subunits by phosphorylating PJA2, which binds and ubiquitinates these subunits, leading to their proteolysis. Additionally, PRKACB phosphorylates GPKOW, regulating its RNA-binding ability. By similar function (by similar): PRKACB also acts as a negative regulator of mTORC1 by mediating phosphorylation of RPTOR.
The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 0.025 nM, the conversion rate is about 25%.
