Background
PUS7 is a pseudouridylate synthase that catalyzes pseudouridylation of RNAs. It regulates protein synthesis in embryonic stem cells by mediating pseudouridylation of tRNA fragments (tRFs), where pseudouridylated tRFs inhibit translation by targeting the translation initiation complex. Additionally, PUS7 catalyzes pseudouridylation of mRNAs with the consensus sequence 5'-UGUAG-3'. It also regulates pre-mRNA splicing by mediating pseudouridylation at alternative splicing regions, with modifications near splice sites directly affecting mRNA splicing and 3'-end processing. Beyond mRNAs and tRNAs, PUS7 binds other RNA types such as snRNAs, Y RNAs, and vault RNAs, suggesting broad RNA pseudouridylation activity. by similar: Pseudouridylation plays a versatile role in RNA function regulation.
