Background
Rad51 plays a crucial role in homologous strand exchange, a key step in DNA repair via homologous recombination (HR). It binds to single-stranded DNA in an ATP-dependent manner to form nucleoprotein filaments essential for homology search and strand exchange. Rad51 catalyzes the recognition of homology and strand exchange between homologous DNA partners, forming a joint molecule between broken DNA and the repair template. During replication stress, Rad51 is recruited to resolve stalled replication forks. Additionally, Rad51 is part of a PALB2-scaffolded HR complex containing BRCA2 and RAD51C, which is thought to function in DNA repair through HR. Under oxidative stress conditions, Rad51, along with RAD51C and XRCC3, regulates mitochondrial DNA copy number. Rad51 is also involved in interstrand cross-link repair.
