RBM38 is an RNA-binding protein that specifically binds to the 3'-UTR of CDKN1A transcripts to maintain their stability, thereby acting as a mediator of the p53/TP53 family to regulate CDKN1A. CDKN1A, a cyclin-dependent kinase inhibitor transcriptionally regulated by the p53/TP53 family, induces cell cycle arrest. Isoform 1 (but not isoform 2) can induce G1-phase cell cycle arrest and stabilize p53/TP53-induced CDKN1A transcripts. Additionally, RBM38 functions as an mRNA splicing factor, specifically regulating the expression of the epithelial cell-specific isoform FGFR2-IIIb, and plays a role in myogenic differentiation.
