Background

REG-3 alpha/REG3A Protein is a bactericidal C-type lectin that lacks the EPN motif, which may explain its inability to bind peptidoglycan. It functions as a hormone in response to various stimuli, including anti-inflammatory signals like IL17A and the gut microbiome. REG-3 alpha/REG3A protein is secreted by different cell types and activates its receptor EXTL3, initiating cell-specific signaling pathways. In keratinocytes, it is induced by IL17A and plays a role in regulating keratinocyte proliferation and differentiation following skin injury. It also inhibits skin inflammation by suppressing inflammatory cytokines like IL6 and TNF. In the pancreas, REG-3 alpha/REG3A protein is able to permeabilize beta-cell membranes and stimulate their proliferation.
