Background
RPS6KA4 is a serine/threonine-protein kinase involved in key biological processes. It phosphorylates CREB1 and ATF1 in response to mitogenic or stress stimuli (e.g., UV-C irradiation, EGF, and anisomycin) and regulates RELA transcriptional activity, particularly in TNF signaling. This kinase also phosphorylates histone H3 at Ser-10 (and potentially Ser-28), activating transcription of immediate early genes like c-fos/FOS and c-jun/JUN. In LPS-stimulated macrophages, it limits pro-inflammatory cytokine production downstream of TLR4, likely by inducing DUSP1 and anti-inflammatory IL10 expression. by similar: Additional studies indicate its role in phosphorylating HMGN1/HMG14 to modulate chromatin structure.
