Background

The SLC19A1 protein operates as an antiporter, facilitating the import of reduced folates or a subset of cyclic dinucleotides through a mechanism driven by the export of organic anions. Functioning as a secondary active transporter, it exports intracellular organic anions down their concentration gradients to facilitate the uptake of its substrates. The protein exhibits high affinity for N5-methyltetrahydrofolate, the predominant circulating form of folate, and can mediate the import of the antifolate drug methotrexate. Additionally, it serves as an importer for immunoreactive cyclic dinucleotides, including cyclic GMP-AMP (2'-3'-cGAMP), an immune messenger produced in response to DNA viruses in the cytosol, and its linkage isomer 3'-3'-cGAMP, playing a role in triggering larger immune responses. Furthermore, the organic anion 5-amino-4-imidazolecarboxamide riboside (AICAR), when phosphorylated to AICAR monophosphate, serves as a substrate for the antiporter activity of SLC19A1.
