It functions as a monomer in synaptic vesicle exocytosis, enhancing processes such as vesicle priming, fusion, and dilation of exocytotic fusion pores. Additionally, SNCA acts as a molecular chaperone when in its multimeric membrane-bound state, assisting in the folding of synaptic fusion components known as SNAREs at the presynaptic plasma membrane. This chaperone activity is vital for maintaining normal SNARE-complex assembly during aging. SNCA also contributes to the regulation of dopamine neurotransmission by interacting with the dopamine transporter (DAT1) and modulating its activity. Existing as both a soluble monomer and homotetramer, SNCA dynamically balances tetramers and monomers, with the tetramer playing a crucial role in homeostasis.
