Background
TPX2 is a spindle assembly factor essential for the normal assembly of mitotic spindles. It is required for proper microtubule assembly during apoptosis and mediates chromatin- and/or kinetochore-dependent microtubule nucleation. TPX2 localizes AURKA to spindle microtubules and activates it by promoting autophosphorylation at Thr-288 while protecting this residue from dephosphorylation. TPX2 is inactivated upon binding to importin-alpha. At the onset of mitosis, GOLGA2 interacts with importin-alpha, releasing TPX2 to activate AURKA kinase and stimulate local microtubule nucleation.
