Background

USP3 is a deubiquitinase involved in various cellular processes, including transcriptional regulation, cell cycle progression, and innate immunity. In response to DNA damage, USP3 deubiquitinates monoubiquitinated target proteins such as histone H2A and H2AX, counteracting RNF168- and RNF8-mediated ubiquitination and facilitating the recruitment of DNA damage repair factors to DNA break sites. It is also essential for proper S-phase progression and mitotic entry, and promotes normal cell proliferation and transformation by deubiquitinating and stabilizing TP53. In innate immune memory establishment, USP3 negatively regulates TLR-induced NF-kappa-B signaling by targeting and removing 'Lys-63'-linked polyubiquitin chains on MYD88. Additionally, USP3 suppresses type I interferon signaling by mediating 'Lys-63'-linked polyubiquitin chains on RIGI and IFIH1. Furthermore, USP3 deubiquitinates ASC/PYCARD, the central adapter for inflammasome assembly and activation, thereby promoting inflammasome activation.

Verified Bioactivity

The fundamental role of USP3 is specific removal of ubiquitin from substrates. USP3 catalyses the ubiquitin from the substrate Ub-Rho110 to release fluorophores. Rho110 will release 535nM emission light under the excitation condition of 485nM. The signal of which can be quickly and reliably captured using a microplate reader. The concentration of USP3 used is 1.5 nM, and the reaction lasts for 30 minutes. Under these conditions, the signal-to-noise ratio (S/N) of the signal box is greater than or equal to 10.
