Background
USP52 is the catalytic subunit of the poly(A)-nuclease (PAN) deadenylation complex, one of two cytoplasmic mRNA deadenylases involved in general and miRNA-mediated mRNA turnover. It specifically shortens poly(A) tails of RNA, with its activity stimulated by poly(A)-binding protein (PABP). Following PAN deadenylation, the CCR4-NOT complex rapidly degrades the shortened mRNA tails. Deadenylated mRNAs are then degraded via two alternative mechanisms: exosome-mediated 3'-5' exonucleolytic degradation or deadenylation-dependent mRNA decapping followed by 5'-3' exonucleolytic degradation by XRN1. Additionally, USP52 acts as a key regulator of the HIF1A-mediated hypoxic response and is required for HIF1A mRNA stability independently of poly(A) tail length regulation.
