(S)-2-Benzylpyrrolidine hydrochloride
(S)-2-Benzylpyrrolidine hydrochloride (Compound s1) serves as a METH hapten scaffold. (S)-2-Benzylpyrrolidine hydrochloride attenuates Methamphetamine-induced hyperlocomotion, hyperthermia and respiratory depression. (S)-2-Benzylpyrrolidine hydrochloride alleviates sedative agent-induced respiratory depression.

Biological Activity
Description
In Vitro
(S)-2-Benzylpyrrolidine hydrochloride has its binding to immobilized H2-BSA inhibited by serum antibodies from H2-KLH vaccinated mice with IC50 values ranging from 3.04 μM to 4.87 μM across 1st, 2nd, and 3rd bleeds[1].

-
Animal Model:Swiss Webster (female, 6 weeks old at study start)[1]
-
Dosage:2 mg/kg; 5 mg/kg; 10 mg/kg
-
Administration:i.p.; single dose
-
Result:Produced locomotor activity comparable to control (no hyperlocomotion) at 2 mg/kg.
Produced dose-dependent suppression of locomotor activity (hypolocomotive effect) at 5 mg/kg and 10 mg/kg.
Significantly attenuated methamphetamine-induced hyperlocomotion when co-administered with 2 mg/kg methamphetamine at 10 mg/kg.
Induced a modest transient 0.6 °C core body temperature increase followed by a 1.5 °C drop below baseline at 10 mg/kg alone; partially reversed methamphetamine-induced hyperthermia after 75 minutes when co-administered with 10 mg/kg methamphetamine at 10 mg/kg.
Increased minute volume (respiratory rate above baseline) at 10 mg/kg alone; reversed methamphetamine-induced respiratory depression and elevated respiration above baseline when co-administered with 2 mg/kg methamphetamine at 10 mg/kg; attenuated fentanyl-induced respiratory suppression, partially restoring normal respiration when co-administered with 0.5 mg/kg fentanyl at 10 mg/kg.
